The pitha karappan is a type of karappan and these symptoms are probably similar to “ Atopic Dermatitis” mentioned in contemporary modern literatures. As per the siddha texts, treatment of pitha Karappan can be effectively done by using various traditional Siddha poly herbal preparations. So this study is aimed to evaluate the Siddha formulations of Karappan Chooranam (KC) is mentioned in the Sarabenthira Vaidhya Muraigal, Karappan Viranaroga Sikichai for the management of Pitha Karappan. The aim of the study is to evaluate the anti-histamine action of the test drug Karappan Chooranam. The study protocol was approved by the Institutional Animal Ethical Committee, S. A Raja’s College of pharmacy, Raja Nagar, Vadakkangulam, Nagarkovil, Tamilnadu, India and anti histamine study was done in vivo animal ( swiss albino mice ) experiment. Bar test was used to study effect of extracts on Clonidine induced catalepsy to determine indirect antihistamine activity. Mice were randomly divided into five groups; First group consist of 3 animals and other four groups consisting of four animals. Group of animals were pretreated with KC at doses ( 100, 200, 300 mg/kg ) and chlorphenaramine maleate ( 10 mg/kg ) and receive Clonidine ( 1 ml/kg ) 30 minutes after treatment. The maximum catalepsy ( 169 0.21 ) was observed in 90 min of clonidine administration ( 1mg/kg ) in control group. The KC at doses low, mid and high mg/kg and chlorpheniramine maleate at 10 mg/kg, showed significant ( p < 0.001 ) These study concluded that the Karappan Chooranam Possess Anti Histamine activity. en_US
